In this experiment, we are doing quantitative determination of Caffeine by reversed-phase high pressure liquid chromatography (HPLC). Elevated column temperature was employed in this method. What impact does increasing column temperature have on retention? On resolution?
Answers
As column temperature increased peak retantion times are shortened as higher temperature leads to faster exchange of analytes between the mobile phase and stationary phase.
As temperature increased resolution decreased as chances of close peaks may get merge.
